RNAiso Blood is a reagent for efficient and rapid RNA isolation from human or animal blood samples or those with high water content. The process involves adding RNAiso Blood solution to disrupt cells, followed by chloroform. After mixing and centrifugation, the solution separates into three layers: RNA in the upper transparent water layer, DNA in the middle layer, and proteins, polysaccharides, fatty acids, and cell debris in the red organic solvent layer at the bottom. The RNA is collected from the upper layer and recovered using isopropanol precipitation. The extraction process is completed in about 1 hour, and the RNA is suitable for applications like RT-PCR, Northern blotting, mRNA separation, and in vitro translation. The red organic solvent layer makes it easy to distinguish the layers. For RT-PCR, it’s recommended to treat RNA with Recombinant DNase I (RNase-free) to eliminate any gDNA contamination that could affect results.
